Adaptive optics for high-resolution fluorescence microscopy

The past quarter century has witnessed rapid developments of fluorescence microscopy techniques that enable structural and functional imaging of biological specimens at unprecedented depth and resolution. The performance of these methods in multicellular organisms, however, is degraded by sample-induced optical aberrations. Here I review recent work on utilizing adaptive optics to improve image quality of fluorescence microscopy for biological samples.

Live demo from the VLT

During the school participants have the exciting opportunity to see a demonstration of the European Southern Observatory’s Very Large Telescope at Cerro Paranal in Chile by Dr Johann Kolb. If conditions allow, this will be performed live.

 Johann Kolb's biography

Johann Kolb is the Adaptive Optics Paranal System Engineer, working on improving the efficiency and performance of the adaptive optics instruments on the VLT but also at La Silla Observatory and maybe one day for the ELT. He completed his PhD between ONERA (France) and ESO Garching (Germany) on “Calibration of ESO’s Multi-Conjugate AO Demonstrator MAD”, followed by 10 more years working on the calibration strategy and system tests of the AO Facility instruments GRAAL (SCAO, GLAO) and GALACSI (GLAO, LTAO). He moved to Chile in 2016 to commission the AO Facility, and then stayed as Adaptive Optics Paranal System Engineer, proposing obsolescence correction and improvements to the current 15 AO systems in operation, with 6 more to come in Paranal and one in La Silla.
